HYPOXIA ASSAY ON THE CYRIS® ANALYSIS PLATFORM

Abstract:
The following exemplary protocol describes the preparation, run, and results of a hypoxia assay with adherent cells of the cell lines L929, HepG2 and J774A.1 in the CYRIS analysis platform. Oxygen (O2) is essential for the entire organism, but also at the cellular level. The cellular respiration can only function through the provision of sufficient oxygen. In the event of hypoxia, cells switch from aerobic energy production to fermentation to generate energy under anaerobic conditions. The hypoxia assay is utilized to create hypoxic conditions and track the cellular adaption to the reduced oxygen supply by measuring the oxygen consumption rate (OCR) and extracellular acidification rate (ECAR) of cells. Here we show the procedure with different cell lines (L929, HepG2 and J447A.1). It is possible and may be necessary to adapt the protocol to other cell types, cell numbers, and culture times.
